Abstract: | Here I describe about the phosphatidylation of functional compounds using phospholipases which is covered with my doctoral thesis. Physiologically active compound such as conjugated fatty acid, terpenes and phenolic compounds sometime exert their activity not fully because of their poor solubility in a certain environment. On the other hand, phospholipids have characteristic amphiphilic property and biocompatibility. In this study, several physiologically active compounds were phosphatidylated using phospholipases such as phospholipase A2 and phospholipase D. The enzymatic reaction was modified and optimized for the yield. In addition, the phosphatidylated compounds were tested for their physiological activity in vitro. Conjugated linoleic acids which have anti-obesity effects were phosphatidylated mediated by phospholipase A2. In this reaction, it is fund that water mimics such as formamide was key factor for the synthetic reaction. Several terpenes and phenolic compounds which have anti-cancer effects or anti-oxidative effects were phosphatidylated mediated by phospholipase D. Some phosphatidylated compounds were exerted higher anti-cancer effects compare to parent compounds. |
